PURPOSE:To provide the titled gene separated by a gene recombination DNA technique and useful for the microbial production of recombinant human cytochrome C. CONSTITUTION:A DNA sequence coding an amino acid sequence of human cytochrome C protein expressed by the formula (the underlined TTG may be TTA). A host cell (e.g. yeast) is transformed with a plasmid containing said DNA sequence and the obtained recombinant host cell is cultured. A recombinant human cytochrome C free from acetylated N-terminal is separated from the cultured bacterial cell and, as necessary, the N-terminal is acetylated to obtain a human cytochrome C useful as a tissue respiration activator. Said DNA sequence can be prepared by conventional process from the whole DNA prepared from a human tissue. As an alternative, it can be prepared by screening a human tissue-originated chromosome DNA library using a 3'-nondecoded region of human cytochrome C cDNA as a probe. |
