<p>PURPOSE:To perform testing large intestine cancer without diet restriction by feces occult blood test, by using a monoclonal antibody KM-371, which belongs to an IgG1 iso type and specifically reacts with human hemoglobin. CONSTITUTION:Hemoglobin of animals, whose meat is provided for meal, such as cattle pigs, chickens and the like is given to a newly born baby mouse within 24hr after the birth. Thus the mouse having immunological tolerance against the hemoglobin is prepared. After the mouse has fully grown up, the mouse is made to have immunity against the human hemoglobin. The splenic cell of the immunized mouse is prepared. The splenic cell and a myeloma cell are fused, and a hybridoma is obtained. The hybridoma is incubated, and a monoclonal antibody KM-371, which specifically reacts with the human hemoglobin, is separated and picked up. Since the monoclonal antibody KM-371 is used, diet restriction is not required, and large bowel cancer can be diagnosed by a feces occult blood test of a patient simply and quickly.</p> |
