发明名称 METHOD FOR IDENTIFYING NUCLEIC ACID SEQUENCE
摘要 PURPOSE:To simultaneously solve degradation in fluorescent intensity and dis lodgment of a dye by converting probe DNA to apurinic acid by hydrolysis and bringing this acid into reaction with a leuco dye. CONSTITUTION:A tail is added to the primary probe having a complementary sequence with the desired nucleic acid sequence and a properly selected secondary probe in such a manner that the primary probe and the secondary probe can make the specific conjugation by the oligonucleotide complementary with each other. Hybridization with the nucleic acid of a sample and the primary probe is then executed. The secondary probe is thereafter hydrolyzed so as to make covalent bonding with acridine yellow. The primary probe and the secondary probe are then conjugated with each other. Labeling is next detected.
