PURPOSE:To extremely exactly determine pyruvate, ADP, etc., in a sample, by using pyruvate oxidase substantially containing no lactate oxidase activity. CONSTITUTION:A water soluble pyruvate oxidase having at least <=0.0005% ATPase content, substantially containing no lactate oxidase activity and capable of catalyzing reaction producing acetyl phosphoric acid, carbon dioxide and hydrogen peroxide from pyruvate, phosphoric acid and oxygen is obtained from a host containing basic sequence coding amino acid sequence of polypeptide containing pyruvate oxidase as a constituent and transformed as an extraneous basic sequence. Then an analyzing composition containing the above-mentioned pyruvate oxidase as an effective ingredient is prepared and pyruvate, ADP, ATP, nonphosphoric acid compound, etc., in a sample is analyzed by using the composition.
