NOVEL ANTIBODY AND DETECTION OF HUMAN HEMOGLOBIN USING SAME

NEW MATERIAL:Anti-human HbmAb capable of selectively bonding to human Hb and hybridoma cell-line. USE:A reagent composed of anti-HbmAb and a test membrane containing said antibody is useful for the test of occult blood. The reagent enables accurate examination with simple operation without necessitating restriction in diet. PREPARATION:A myeloma cell is fused with an antibody-producing cell produced by immunizing an animal other than human with human Hb as an antigen. The obtained hybridoma is cultured in a medium or transplanted into peritoneal cavity of mouse to form ascites and obtain anti-HbmAb capable of selectively bonding to human Hb. The antibody is supported on a membrane composed of a polymer containing introduced epoxy group, etc., to obtain an antibody-bonded membrane. A determination kit is prepared by combining said membrane with an indicator-containing membrane or using an antibody- bonded membrane containing color-developing agent.
