PURPOSE:To obtain a phospholipid containing two phosphatidyl groups, by reacting a specific phospholipid with a chain polyhydric alcohol in the presence of phospholipase having phosphatidyl group rearranging activity. CONSTITUTION:A raw material shown by formula I [R1 and R2 are acyl or alkyl; R3 is alkylene which may be substituted with OH, alkoxyl or hydroxyalkyl; X is organic group obtained by removing one hydroxyl group from hydroxyl group-containing base] is suspended in water, is reacted with about 2-40mol. based on phospholipid of a polyhydric alcohol shown by formula II in the presence of phospholipase D (EC3.1.4.4) derived from a ray fungus and 4-30mM calcium ion at 20-60 deg.C, preferably 35-50 deg.C at pH >=4, preferably pH 5-8.4 and a phospholipid shown by formula III is isolated from the reaction mixture. 1-2% organic solvent such as ethyl acetate may be added to the reaction system to promote dispersion of phospholipid and to raise reaction efficiency.
