PURPOSE:To determine a substrate or enzyme with a high accuracy, by reacting the substrate, etc., with H2O2 and an oxidase capable of forming superoxide anions and measuring the H2O2 contained in the reaction system using peroxidase based coloring reaction, etc., after completing the above-mentioned reaction. CONSTITUTION:A substrate (e.g. uric acid) or a substrate (e.g. creatinine) consisting of a substance formed by enzymic reaction is reacted with hydrogen peroxide and an oxidase capable of forming superoxide anions (e.g. uricase) to carry out reaction. After completing the oxidase reaction and converting the formed superoxide anions into hydrogen peroxide, the hydrogen peroxide contained in the reaction system is measured by peroxide based coloring reaction or fluorescent emitting reaction to determine the substrate or enzymic activity by utilizing the oxidase reaction.
