NEW MATERIAL:A colony stimulating factor, which is a homodimer, consisting of two same subunits having an amino acid sequence expressed by then formula and having a colony formation stimulating action on mammalian monocytic-macrophage based cells and the following properties. Molecular weight measured by a sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (PAGE) method; 70,000-90,000 daltons and 35,000-45,000 daltons for subunits treated with a reducing agent. Isoelectric point (pI); 3.1-3.7. Containing saccharide chains, having alpha-helical structure without losing biological activity even by heating at 60+ or -0.5 deg.C for 60min. USE:A remedy for leukopenia, immunodeficiency, bone marrow transplantation, etc. PREPARATION:Human urine is adjusted to pH 8-9 and insoluble substances are precipitated and separated. The resultant urine is concentrated and adjusted to pH 6.5-7.5, heat-treated at 60 deg.C and adsorbed on an anion exchanger. The adsorbed substance is then eluted and subjected to gel filtration to recover a fraction having >=70,000 molecular weight, which is subsequently adsorbed on a hydrophobic compatible substance, eluted and subjected to high-speed liquid chromatography to afford the aimed substance having subunits expressed by the formula.
