PURPOSE:To permit the large-volume production of pullulanase in an appropriate host by determination of the base sequence of the pullulanase gene and the amino acid sequences of precursor protein and mature protein of pullulanase. CONSTITUTION:The pullulanase gene has been identified as a base sequence of formula I (base number reaches 3231 but its detail is omitted) and a biologically purified base sequence bioequivalent to formula I. In formula I, X is 0 (no thing) or a polynucleotide with the base sequence given in formula II. Further, pullulanase having the amino acid sequence of formula III (the amino acid number reaches 1077, but the detail is omitted) or the amino acid sequence substantially bioequivalent to formula III has been determined. In the formula III, Y represents H atom or formula IV. Further, when Y is H atom, the Cys on the N-terminal may be modified with palmitic acid. |
