<p>PURPOSE:To obtain a readily producible human amyloid-related protein monoclonal antibody, capable of specifically recognizing a human amyloid- related protein and useful for diagnosing Alzheimer type dementia, etc. CONSTITUTION:Mice, etc., are immunized with a human amyloid protein obtained by extracting an autoptic material, such as a patient suffering from amyloidosis, and spleens, etc., are excised to provide antibody-producing cells (A), which are then mixed with myelomatous cells, such as myeloma cells of mice, at 5-10:1 ratio. A fusion promoter [e.g. polyethylene glycol (PEG)] is added to carry out cell fusion and afford fused cells (B), which are subsequently selected and cultivated to provide a hybridoma. The resultant hybridoma is then cloned to afford a monoclonal hybridoma (C), which is subsequently cultivated in an Eagle culture medium, etc., for 3-5 days. The obtained culture medium supernatant is dialyzed and purified to collect the aimed human amyloid-related protein monoclonal antibody having about 42 or 45 kilodaltons molecular weight measured by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (PAGE) and a western blotting method.</p> |
