Prepn. of L-glutamic acid by fed-batch culture of Brev. lactofermentum or C. glutamicum on the excess biotin-contg. medium with controlled sugar and antifoamer feeding comprises inoculating the seed culture of C. glutamicum ATCC 13032 into a 30 liter fermentor contg. 13 liter medium (pH 7.6) consisting 8.0 sugar beet molasses, 0.1 K2HPO4, 0.05 MgSO4.7H2O, 0.00001 thiamine.HCL, 0.3 corn steep liquor and 0.02-0.01% w/v polyoxyalkylene antifoamer (HLB value, 2.0-4.0) and culturing with feeding 0.05-0.3% w/v (based on fermentation broth) antifoamer mixt. of 60-80% polyoxyalkylene gp. (HLB, 2.0-4.0) and 20-40% polyoxyalkylene gp. (HLB, 5.0-9.0) by wt.
