PURPOSE:To obtain a high-purity saccharide (derivative) without forming a rearrangement product, by reacting a saccharide or free hydroxyl group- containing saccharide derivative with a phosphinothioyl halide and protecting the saccharide hydroxyl groups with phosphinothioyl groups. CONSTITUTION:A saccharide or free hydroxyl group-containing saccharide is initially reacted with a phosphinothioyl halide expressed by the formula (X is halogen; R is alkyl or aryl) (preferably dimethylphosphinothioyl chloride) in the presence of a base (preferably n-butyllithium) to provide a phosphinothioylated saccharide derivative, which is then reacted with hydroxide ions (preferably trimethylbenzylammonium hydroxide) to remove the phosphinothioyl group and afford a saccharide (derivative). Furthermore, phosphinothioylation of the raw material saccharide (derivative) is preferably carried out at a reaction temperature of 0 deg.C - room temperature using dichloromethane as a solvent.
