PURPOSE:To collect thermostable aminoacylase SK-2 in high yield, by cultivating a strain such as Bacillus thermoglucosidius belonging to the genus Bacillus and forming thermostable aminoacylase SK-2 in a culture mixture. CONSTITUTION:A strain such as Bacillus thermoglucoxydas belonging to the genus Bacillus, capable of producing thermostable aminoacylase SK-2, is cultivated, thermostable aminoacylase SK-2 is formed in the culture mixture and thermostable aminoacylase SK-2 is collected from the culture mixture. The thermostable aminoacylase SK-2 acts on (1) N-acetyl-D,L.amino acid to liberate L-amino acid, acts only on (2) L-form of acetylated amino acid but not on D-form at all. In the acetylated amino acid, the thermostable aminoacylase SK-2 acts on N-acetyl-L-methionine and N-acetyl-L-alanine approximately in the same degree.
