PURPOSE:To establish the method of a highly sensitive quantitative determination excellent in specificity and reproducibility and having a short measuring time by using a specific monoclonal anti-PA (human prostate specific antigen) antibody and a specific polyclonal anti-PA antibody in combination. CONSTITUTION:In the method of the quantitative determination of a PA by sandwich type immunological measuring method, a monoclonal anti-PA antibody having specificity in the PA produced by a hybridoma formed by the fusion of a cell from the tumor line of a mouse and a spleen cell from a mouse immunized by the PA in advance and a polyclonal anti-PA antibody obtained from an animal immunized by the PA are used in combination. It is left to discretion to determine whether the monoclonal antibody or the polyclonal antibody is selected as an immobilized antibody or a labeled antibody. However, from the viewpoints of sensitivity, specificity and reproducibility, it is preferable that the monoclonal antibody is made an immobilized antibody wherein the monoclonal antibody is fixed to a water-insoluble carrier, such as a glass bead, and the polyclonal antibody wherein a rabbit anti-PA antibody (class: IgG), after rendered Fab', is labeled with an enzyme by maleimide method is made a labeled antibody. |
