Sisomicin, useful as an antibactericide, is prepd. by cultivation of a mutant (KFCC 10188), which is derived from Micromonospora inyoensis ATCC 27600 by UV mutagenesis, can grow on high sisomicin and is resistant to methionine and CoCl2, on a nutrient medium contg. ethionine and CoCl2 under submerged aerobic conditions. Thus, the 2nd stage precultures of a mutant was inoculated into a 30 liter fermentor contg. 14.25 liter medium consisting of dextrin 70, soybean cake 35, CaCo35, corn steep liquor 5, methionine 2 and CoCl2 0.005g per litre distld. water and cultivated for 6 days at 32≦̸C and pH 7.0 under aeration (1.5vvm) and agitation (200rpm) to give 1870 μg sisomicin per ml culture broth.
