PURPOSE:To enable accurate determination of alpha-amylase activity in a specimen, by contacting a specimen with a substrate containing maltooligosaccharide in the presence of glucosidase and contacting isolated inositol with an inositol dehydrogenase. CONSTITUTION:A specimen is made to contact with a substrate containing a maltooligosaccharide of formula I [A is group of formula II (R1-R4 are H, etc.; X1-X4 are O, etc.), etc.; I is inositol residue, etc.; G is glucose residue; n is integer of 3-15] in the presence of glucosidase to isolate inositol or its derivative. The inositol is made to contact with inositol dehydrogenase in the presence of NAD to produce NADH. The change in absorbance of the NADH is measured to determine the amount of inositol, from which alpha-amylase activity can be calculated.
