发明名称 METHOD FOR DETERMINING BASE SEQUENCE
摘要 PURPOSE:To make detection at high S/N and to decrease an error rate by using a phosphorescent material as a labeling dye and converting excitation light to square continuous pulse light of about <=1msec rise time as an optical method for detecting phosphorescence. CONSTITUTION:DNA fragments labeled with EITC are injected into sample injection ports 8 according to bases A, T, G, C at the terminal and a voltage is impressed between the electrolytes of electrode cells 4, 6 from a power source 10 for migration to migrate the respective samples along a slab gel 2. Laser light from an argon laser is then converted to pulses by a shutter 14 and these pulses are projected as the excitation light 18 from a top end 30 of a photometric system through half mirrors 16, 22, a lens 24 an optical fiber 26 to a gel 2. While the top end 30 is moved in an arrow direction, the phosphorescence emitted from the DNA fragments migrated by the gel 2 is collected by an objective lens 28 at the top end 30 and is entered to a fiber 26. Said light is entered through the fiber 26, a lens 24 and a mirror 22 to an interference filter 34. The light is collected by a lens 36 and is entered to a photoelectric multiplier 40, by which the detection is executed.
