PURPOSE:To quickly and surely detect the prescribed components in a specimen by using a reversed-phase column as a stationary phase and a phosphoric acid buffer soln. which is dissolved with prescribed components at prescribed ratios and is adjusted in pH to 2-4 as a mobile phase. CONSTITUTION:One end of the reversed-phase column 1 in which the stationary phase formed by chemically bonding an octadecyl group to the surface of silica particles is connected via a sample injection port 3 to a mobile phase tank 2 and the other end is connected via a branch pipe 4 to a derivatization reagent tank 5 and a UV fluorescent detector 6. Liquid feed pumps 7, 8 are provided to said column. An aq. soln. of 5-10mol. phosphoric acid contg. 20-30mol. sodium pentasulfonate is housed into said mobile phase tank 2 and an orthphthalic aldehyde soln. is housed into the reagent tank 5. Histidines form ion pairs selectively with the sodium pentasulfonate in the mobile phase and said pairs are held by reversed-phase ion pair chromatography. As a result, the histidines are eluted from the column in order of 1-methyl histidine, histidine and 3-methyl histidine. The sepn. and detection of the histidines in about 1/5 shorter time than heretofore is thus enabled.
