PURPOSE:To prepare a reagent composition having a specific diol group obtained by reacting a biologically active substance having amino group with alpha- acetylmercaptosuccinic anhydride and use the substance instead of each ingredient. CONSTITUTION:A compound expressed by formula I (R1 and R2 are hetero ring or aromatic ring residues exhibiting different s-s exchange reactivity) is reacted with a compound having thiol group expressed by the formula A1-SH in an inert solvent such as water, buffer, dimethylformamide, etc., at 10-40 deg.C for 10min-40hr. The reaction product is recovered and purified by dissolution, solid-liquid separation, etc., to provide the aimed reagent composition expressed by formula II (A1 and A2 are biologically active substances, supports or thiol residues or reactive groups of thiolated modifying derivative thereof; X is spacer group).
