DETERMINATION OF N-ACETYLNEURAMINIC ACID AND KIT THEREFOR

PURPOSE:To determine the titled compound for diagnosis of inflammation, etc., without an influence of endogenous pyruvic acid, by reacting a sample with N-acetylneuraminic aldolase and N-acetylneuraminic dehydrogenase and measuring the resultant reduced form nicotinamide adenine dinucleotide (NADH). CONSTITUTION:N-Acetylneuraminic aldolase and N-acetylmannosamine dehydrogenase are successively or simultaneously reacted with an N- acetylneuraminic acid-containing sample to form reduced form nicotinamide adenine dinucleotide (NADH), which is measured by absorbance at 340nm wavelength. The concentration is then obtained from a calibration curve prepared by measuring N-acetylneuraminic acid solutions in known concentrations by the similar method to determine the N-acetylneuraminic acid in the sample.
