PURPOSE:To obtain a vector, containing a DNA sequence prepared by inserting a chloramphenicol acetyltransferase (CAT) structural gene into a plasmid pTT8 of Thermus thermophilus, selective by chloramphenicol and useful for a highly thermophilic microorganism. CONSTITUTION:A plasmid pC194 is treated with a restriction enzyme Bal31 to provide a fragment containing CAT (chloramphenicol acetyltransferase), which is then linked to a Bal31 fragment containing a promoter of a plasmid pTT8 by ligase treatment and introduced into a highly thermophilic microorganism to select a chloramphenicol-resistant colony and provide a chloramphenicol- resistant plasmid. Alternatively, an Escherichia coli plasmid pBR322 is inserted into a plasmid pTT8 to prepare a shuttle plasmid, which can be used as a material, linked to a CAT gene and introduced into Escherichia coli to afford a drug-resistant plasmid. Thereby a vector usable in the genus Thermus is prepared from the above-mentioned plasmid. |
