PURPOSE:To obtain purified myoglobin in high purity, by treating crude myoglobin by a combination of ammonium sulfate fractionation, gel-filtration and ion-exchange chromatography. CONSTITUTION:A crude myoglobin obtained by the centrifugal precipitation, etc., of myoglobin urine, myoglobin serum, homogenate of skeletal muscle, etc., is used as a raw material and subjected to salting-out with ammonium sulfate. The precipitate is further salted out with ammonium sulfate and fractionated stepwise by successively using ammonium sulfate having increasing concentration. The obtained precipitate is dissolved in water and the solution is subjected to gel-filtration. The objective purified myoglobin is produced by carrying out ion-exchange chromatography of the gel-filtration product. |
