Liposomes encapsulating a labeling substance, and streptolysin O (SLO) are added to a serum sample, and after the antigen-antibody reaction between antistreptolysin O (ASO) in the sample and SLO added, the amount of the labeling substance released by lysis of the liposome membrane by the residual SLO is measured, whereby ASO value in the sample is determined. According to this determination method, ASO value can be determined rapidly and accurately and the determination can be automated.
