PURPOSE:To obtain the titled analysis element having improved preservability and capable of rapidly determining a small amount of specimen, by placing a glycerol-detecting color-developing reagent layer to one surface of a water- impermeable substrate and adding a lipoprotein lipase and an alkaline pH- buffering agent to a developing layer composed of a woven fabric, etc. CONSTITUTION:A glycerol-detecting color-developing reagent layer is formed on a water-impermeable substrate by applying an aqueous solution containing a glycerol-detecting color-developing reagent composition and a hydrophilic polymer binder and having 6-7.5pH to the substrate and drying the solution. The glycerol-detecting color-developing reagent layer is produced by applying an aqueous solution having 6-7.5pH and at least containing glycerol kinase, glycerol-3-phosphoric acid dehydrogenase, an electron-transfer compound, NAD, ATP, a tetrazolium salt and magnesium ion. A porous developing layer composed of a woven fabric, etc., is directly or indirectly formed on said reagent layer and a liquid containing dissolved lipoprotein, a buffering agent and an alkali agent is applied to the porous layer and dried to obtain the objective analysis element.
