PURPOSE:To permit selective oxidation and easy determination of direct type bilirubin by adding copper ion and thiourea or the deriv. thereof to a liquid to be inspected. CONSTITUTION:Absorbancy of light of the liquid to be inspected at about 450nm is previously measured and the copper salt and thiourea or the deriv. thereof is added thereto. The absorption of the reaction liquid at about 450nm is then measured after the end of the reaction of the direct type bilirubin and the copper ion and thiourea or the deriv. thereof. The concn. of the direct type bilirubin is determined from the difference in the absorption before and after the reaction. This measurement is based on that the copper ion and thiourea or the deriv. thereof forms a complex and the complex exhibits the effect of selectively oxidizing the direct type bilirubin. The pH of the reaction liquid is preferably an acid side, more particularly preferably <=6 in order for the copper salt to exist in the form of copper ions in the reaction liquid. The direct type bilirubin is selectively oxidized and is easily determined in the above- mentioned manner.
