PURPOSE:To carry out preparation of two enzymes at one time, by preparing a plasmid containing both a gene to code L-alanine dehydrogenase and a gene to code D-amino acid transaminase. CONSTITUTION:A DNA fragment containing a gene to code L-alanine dehydrogenase and a DNA fragment containing a gene to code D-amino acid transaminase are collected, both the collected genes and DNA playing a role as a vector are digested with a restriction enzyme and bonded with a ligase to prepare a recombinant plasmid. The Escherichia coli HB101 treated with calcium chloride at about 0 deg.C is brought into contact with the recombinant plasmid to give transformed Escherichia coli. |
