PURPOSE:To dispense a required amt. of a water layer contg. DNA without detecting the boundary between two layers by assigning the stop position at the time when the top end part of a dispenser sucks liquid according to the injection rate of phenol. CONSTITUTION:The phenol is mixed with a DNA sample and is separated by centrifugal sepn. to the water layer contg. DNA and the phenol layer which are set in a vessel 8 of the dispenser. The stop position of the top end of a dispensing tip 1 is calculated by a controller 10 in accordance with the amt. of the phenol injected into the vessel 8 and a stepping motor 7a is driven to lower the tip 1 down to the stop position. A plunder driving mechanism is then driven to rise a plunger 5 so that the water layer part is sucked and is discharged into a fresh container. Then, the aq. soln. contg. DNA is easily separated from the phenol layer.
