PURPOSE:To determine reduced form nicotinamide adenine dinucleotide (NADH) or reduced form nicotinamide adenine dinucleotide phosphate (NADPH) in high sensitivity, by converting the NADH or NADPH into hydrogen peroxide and measuring the resultant hydrogen peroxide by a chemiluminescent method. CONSTITUTION:At least one of reduced from nicotinamide adenine dinucleotide (phosphate)-flavin mononucleotide [NAD(P)H-FMN] oxidoreductase, NADH- FMN oxidoreductace, NADPH-FMN oxidoreductase, diaphorase and old yellow enzyme is reacted with an NADH or NADPH-containing sample in the presence of flavins to produce reduced form flavins, which are then oxidized to form hydrogen peroxide. An oxidizable nonfluorescent substance is reacted with the resultant hydrogen peroxide through an oxidation catalyst and converted into a fluorescent substance, which is then reacted with succinic diesters and hydrogen peroxide under inhibiting condition of the oxidation catalyst to measure the quantity of emitted light and determine the NADH or NADPH in the sample.
