PURPOSE:To make it possible to transduce DNA into an eukaryocyte simply and efficiently, by bringing DNA as a complex with protamine into contact with a culture cell. CONSTITUTION:DNA such as cyclic or linear plasmid, e.g. psV2-CAT, pAD12E 1A-CAT, psV2-neo, etc., plasmid fragment, chromosome, e.g. Ad2DNA, etc., chromosome fragment, etc., is blended with protamine derived from sperm of salmon, herring, gray mullet, green sturgeon, human, etc., in a ratio of 1:100 in an aqueous medium at pH 7.5-8.5, kept at 9 deg.C for about 30min, then at 30-40 deg.C for about 10min and recooled to 0 deg.C to form a complex. The prepared DNA-protamine complex is once isolated or an aqueous medium containing the complex is directly added to an eukaryocyte containing a cell derived from animal or plant, preferably a culture medium for animal cell and the complex is in contact with the cell for 20min-5hr to transduce DNA into the cell. |
