New bilirubin oxidase M-1 was produced by culturing Trachyderma tsunodae K-2593, purified, and used as analytical reagent. Thus, bilirubin oxidase produced by culturing in medium seeded with T. tsunodae K-2593 and purified by (NH4)2SO4 fractionation, chromatog. on DEAE-Sephadax A-50, DEAE-Sepharose CL-6B, and gel filtration on Sephacryl S-200, was used in a reagent contg., in addn. to the enzyme, Na deoxycholate for the detn. of bilirubin in body fluids. Following incubation with MBTH in citrate buffer, pH5.5, the change in absorbance was measured at 610mm.
