<p>PURPOSE:To produce efficiently L-tryptophan from a carbohydrate, anthranilic acid or indole, by using coryneform bacteria transformed with a recombinant DNA having tryptophan synthetase (TS) gene. CONSTITUTION:A chromosome gene is extracted from a coryneform bacterial strain, e.g. Bravibacterium divaricatum ATCC14020 or Brevibacterium saccharolyticum ATCC14066, and then cleaved with a suitable restriction enzyme. The cleaved fragment is then linked to a plasmid vector, e.g. pAJ1844, and the resultant recombinant DNA is used to transform coryneform bacteria. The resultant transformant is cultivated in a carbohydrate culture medium or a culture medium containing anthranilic acid or indole added thereto to form the aimed L-tryptophan.</p> |
