NOVEL COSMID VECTOR HAVING AT LEAST TWO REPLICATION FUNCTION HAVING DIFFERENT INCOMPATIBILITY GROUP AND PRODUCTION OF GENE BANK USING THE SAME

摘要

PURPOSE:To produce a gene bank of foreign DNA, by producing a cosmid vector having >=two cos of lambda-phage having same directivity and >=two replication functions of different incompatibility groups. CONSTITUTION:A cosmid vector having >=two cos of lambda-phage or lambdoid- phage having same directivity and >=two replication functions of different incompatibility groups is produced beforehand. The cosmid vector is cut with two kinds of restriction enzymes and various foreign DNA fragments having length of 35-45kbp are inserted between the right arm and left arm having at least one cos produced by the above cutting process. The phage is converted to a transduced phage particle by the above in-vitro packaging process. The phage is cloned to E.coli to obtain a gene bank of foreign DNA.
