PURPOSE:To readily produce acetyl-CoA from a coenzyme A and S- acetylglutathione at a low cost, by using glutathione thiol esterase activity- containing substance. CONSTITUTION:Yeast cells belonging to the genus Saccharomyces, Candida, Hansenula, Schizosaccharomyces, Pichia, Kloeckera, etc., is cultivated in a nutrient culture medium to prepare a glutathione thiol ester active fraction from wet yeast cells. The above-mentioned yeast cells are immobilized in a polyacrylamide carrier to give immobilized yeast cells. A glutathione thiol esterase-containing substance, e.g. the above-mentioned active faction, immobilized yeast cells, etc., is blended with preferably 3-6mM coenzyme A and 20-40mM S-acetylglutathione and reacted at 8-9pH and 37 deg.C for 15-30min to afford acetyl-CoA. The immobilized yeast cells are filled in a column and a substrate can be continuously passed therethrough to continuously give the acetyl-CoA. |
