PURPOSE:To readily purify a basic peptide such as secretin family or the like useful as a medicine in large quantities, by extracting the unpurified basic paptide with an acidic and aqueous solution and subjecting the extracted solution to a weak cation-exchange chromatography. CONSTITUTION:A basic peptide such as VIP (Vasoactive Intestinal Polypeptide) or the like belonging to a secretin family synthesized by a solid phase synthetic method, etc., is extracted with an acidic and aqueous solution (e.g. aqueous acetic acid solution, etc.) and then freeze-dried under reduced pressure and water or an aqueous organic acid ammonium salt solution (e.g. aqueous ammonium acetate solution) is added thereto to dissolve the basic peptide and then the basic peptide solution brought into contact with a weak cation-exchange resin (e.g. carboxymethyl cellulose, etc.) to adsorb the basic peptide, which is successively eluted with an organic ammonium salt and as necessary subjected to high speed liquid chromatography to remove a slight amount of impurity. |
