<p>A biphasic culturing apparatus which employs multiple solid nutrient media in a single vessel (10) includes an enclosed container (12) which has an opening (34) defined in one of its surfaces for introducing material into the container. At least one container surface (14, 16, 18, 20) includes a flat, transparent portion for viewing the solid and liquid nutrient media from outside the container. A first partition (22) inside the container divides the interior volume thereof into upper (26) and lower (28) portions, and at least one second partition (24) divides the upper container portion into a plurality of compartmental volumes. A liquid nutrient medium is located in the lower container portion, and a solid nutrient medium is contained in each compartment volume. The first and second partitions are configured so that each solid nutrient medium may be separately inoculated with the liquid medium. To prepare the culture vessel for use, a plurality of preselected culturing base materials are deposited into the compartmental volumes, and the appropriate liquid nutrient medium is deposited into the lower container portion. To achieve sterilization, the base materials and liquid medium may be aseptically deposited into the culture vessel, or the entire assembly may be autoclaved. A method for detecting microorganisms in a specimen includes introducing the specimen into the liquid nutrient medium and subsequently inoculating preselected ones of the solid nutrient media with the liquid medium. After a suitable incubation period, the solid nutrient media are examined for the presence of microorganisms.</p> |
