PURPOSE:To purify human tissue plasminogen activator (TPA for short) in high yield not only form serum-free culture solution but also from serum- containing culture solution, by adsorbing TPA in an adsorbing carrier and eluting TPA with a solution containing omega-amino acid. CONSTITUTION:Human tissue plasminogen activator is adsorbed on an adsorbing carrier and eluted with a solution containing omega-amino acid after adsorption. Controlled pore glass (CPG), silica gel or diatomaceous earth is preferable as the adsorbing carrier. L-Lysine, 4-aminobutanoic acid, 5-aminopentanoic acid, 6-aminohexanoic acid, 7-aminoheptanoic acid, 8-aminooctanoic acid, tranexamic acid, etc., may be cited as the omega-amino acid. The above-mentioned TPA is produced by a cell transformed with TPA manifestation vector. |
