Device and method for detecting and quantifying a substance by filtration. The device comprises a body consisting of two parts (1 and 2) nested into each other, of which one (1) is used as a reservoir and the other (2) is used as filter-holder and as receptacle, and a retention and reaction system consisting of a strut (3), two multiperforated rigid plates (4 and 5) which imprison a filtering membrane (6) and a thin strut (7), and a seal (8). The reservoir (1a) and the receptacle (2a) are connected to an external circuit by means of conduits (1b and 2b), mouthpieces (9 and 10) and pipes (11 and 12). The interlocking of the two parts (1 and 2) of the body is obtained by means of joining hooks (13). The thickness of the strut (3) is determined as a function of the volume of the virtual chamber to be obtained.
