NEW MATERIAL:A compound shown by formula I [B is nucleic acid base residue which may contain protective group; R<1> is OH-protecting group; R<2> is group to be made into phosphoric acid-protecting group; R<3> is (substituted)alkyl]. EXAMPLE:5'-O-Dimethoxytrityl-3-N-benzoylthymidine-3'-O-(S-t-butylthio) methyl phoshorothioite. USE:A base for genetic engineering. PREPARATION:Nucleoside containing protected hydroxyl group at the 5'-position is reacted with an alkylthiochloroalkoxyphosphine (e.g. t- butylthiochloromethoxyphosphine, etc.) shown by formula II.
