<p>PURPOSE:To obtain protoplast of gramineous plant in high yield, by treating a cut piece of leaf of gramineous plant in a specific enzyme solution and dissolving the cell wall. CONSTITUTION:A leaf (preferably young leaf about 4-7 days after germination) of gramineous plant is finely cut (preferably 1-2mm square, treated with an enzyme solution containing endo-1,4-beta-glucanase, xylanase and endo-1,3-beta- glucanase, the cell wall is dissolved and protoplast is separated and collected from the solution. The enzyme solution preferably contains 50-400 units/ml endo-1,4-beta-glucanase, 400-1,800 units/ml xylanase and 50-450 units/ml endo-1,3-gamma-glucanase. The enzyme solution is preferably buffered at pH about 5.0-5.5.</p> |
