PROBLEM TO BE SOLVED: To obtain a new protein for preparing an antibody suitable for analyzing the action of a hepatocyte growth factor(HGF) activator, its assay, etc., inhibiting protease activity of a hepatocyte growth factor(HGF) activator, having a specific amino acid sequence and molecular weight. SOLUTION: This new protein has about 30,000 dalton molecular weight by SDS-polyacrylamide gel electrophoresis, activity of suppressing protease activity of a hepatocyte growth factor(HGF) activator and any of amino acid sequences shown by formulas I to III or a substantially same amino acid sequence and is useful as an activity controlling factor for HGF and HGF activator or is useful for preparing an antibody suitable for physiological action of HGF activator, action analysis and the analysis of the control mechanism of HGF activation. The protein is obtained by culturing a human cancerous cell strain in a serum-free medium, recovering and concentrating the supernatant liquid of the culture product, supplying the concentrated solution to a heparin immobilized column, eluting an adsorbed fraction and fractionating the eluted solution by column chromatography. |
