NEW MATERIAL:An enzyme derived from a bacterium belonging to the genus Clostridium, capable of producing endo-beta-galactosidase. Substrate specificity: acting on a part having Gal alpha1 3Gal beta1 4GlcNAc sequence and cleaving Gal beta1 4GlcNAc bond at the end and not on a part having Gal alpha1 3(Fuc alpha1 2)Gal beta1 4GlcNAc sequence at the end. Optimum pH: 6.5. Stable pH range: activity is halved at pH 5 and pH 8.5. USE:Useful for analyzing saccharide chain structure of conjugated glucide existing in blood type substance of human erythrocyte. PREPARATION:A bacterium [e.g. Clostridium perfingens (FERM P8917)] belonging to the genus Clostridium, capable of endo-beta-galactosidase, is cultivated in a liquid medium by spinner culture method, etc., and the aimed substance is isolated from the culture mixture. The culture is preferably carried out at 37 deg.C for 10-100hr. |
