NEW MATERIAL:Epithelial growth inhibitor (EGI for short) (molecular weight: about 27,000; being deactivated by reduction; consisting of two subunits). USE:A carcinostatic agent inhibiting multiplication of human squamous cell carcinoma cell. PREPARATION:Human blood is centrifuged, erythrocyte is precipitated and supernatant liquid is collected. The supernatant liquid is centrifuged to give blood platelet precipitate, which is suspended in PBS, centrifuged again to give blood platelet precipitate. The precipitate is suspended in acidic ethanol having a composition shown by table, treated by a homogenizer, the prepared homogeneous solution is centrifuged, the precipitate is dried in vacuum, dissolved in 1M acetic acid and the prepared solution is used as an extracted solution of blood platelet. The extracted solution is sent to equilibrated TOYOPAL HW50 column, developed with 1M acetic acid and treated by chromatography to elute a large amount of EGI of fraction number 23-28. The EGI is collected and purified to give almost pure EGI. |
