PURPOSE:To produce catalase, by isolating catalase cDNA using a DNA probe having a specific base sequence chemically synthesized from a cDNA bank prepared from human hepatic cells and introducing the above-mentioned DNA into a host. CONSTITUTION:A cDNA binary of human hepatic cells is prepared by the Kwok et al. method and a DNA, chemically synthesized from the above- mentioned library on the basis of the partial sequence of catalase cDNA and consisting of 33 bases is used to give a cloned pCD-Cat89 having the longest catalase cDNA. Fragments cleaved with BamHI of the above-mentioned clone are subcloned to an NcoI site of pkk233-2 vector to give a plasmid pkk-Cat250, which is used to transform Escherichia coli RB791 and produce the aimed catalase. The above-mentioned plasmid can be similarly introduced into a yeast or animal cell to carry out gene expression. |
