发明名称 IMMOBILIZATION OF LIGAND ON INORGANIC CARRIER
摘要 PURPOSE:To enable the immobilizing of a ligand stably in a wide pH range, by making an imino compound or an amino compound adsorbed on an inorganic carrier to immobilize a ligand thereon through a crosslinking agent. CONSTITUTION:An inorganic carrier mainly composed of SiO2 and Al2O3 or that made of metal oxide such as NiO is washed with water, made to react with an aqueous solution of an imino compound such as polyethylene imine (MW=6,000-8,000) or an amino compound such as gelatin and washed with water to remove access imino compound or amino compound. Then, a crosslinking agent such as glutaric aldehyde is caused to react with the carrier and after washing with water, a functional group such as aldehyde group is introduced thereinto. Furthermore, a ligand such as human IgE monoclonal antibody is made to react with the carrier to be immobilized thereon. The immobilized ligand thus obtained can be used for the measurement of human IgE or the refining of the monoclonal antibody.
