<p>PURPOSE:To extract and separate valuable component of a cell in a short time facilitating the recovery of solvent, by mixing a cultured liquid of microbial cells with a solvent solution of supercritical state, subjecting the mixture to a specific treatment in a separation tank to disintegrate the cell membrane and precipitating and separating the product by gravitational force. CONSTITUTION:A cultured liquid of microbial cell is mixed with a solvent solution of supercritical state or pseudocritical state and the mixture is introduced into a 1st separation tank after instantaneously lowering the pressure. The treated liquid of microbial cell unloaded from the bottom of the 1st separation tank is introduced into an extraction tank and brought into countercurrently contact with a solvent solution of supercritical state or pseudocritical state. A solvent solution phase containing dissolved microbial cell extract is taken out of the extraction tank from its top and introduced into a 2nd separation tank. The solution is subjected to gravitational precipitation and separation in the 2nd separation tank under a pressure condition to separate the solvent solution from the cell extract. The cell extract is recovered from the bottom of the 2nd separation tank and the solvent solution is recovered from its top. The amount of the solvent solution is preferably 1-10pts.wt. per 1pt.wt. of the cultured liquid of microbial cell.</p> |
