PURPOSE:To detect nucleic acid using nucleic acid not labelled with a re dioisotope as a probe, by using modified nucleic acid. CONSTITUTION:Nucleic acid is modified without using a marker at all and modified nucleic acid is used as a probe. At first, the modification of nucleic acid is performed by a physical or chemical means. For example, there are an ultraviolet ray irradiating method and an oxidizing or reducing method. When the modified nucleic acid (a) is two-chain one, it is modified to one-chain nucleic acid. When nucleic acid is treated with a usual method of heat treatment of alkali treatment, one-chain modified nucleic acid (b) is generated. Next, said one-chain modified nucleic acid an the one-chain nucleic acid of a specimen are hybridized and hybrid nucleic acid is obtained. Then, said modified nucleic acid (b) and a substance confirming the change thereof are reacted to obtain a reaction product (e) which is, in turn, measured corresponding to the kind thereof by a usual method.
