FLAVIVIRUS ANTIGEN

摘要

PURPOSE:To provide a flavivirus antigen containing at least one kind of epitope of V3 protein of flavivirus and effectively utilizable as a high-purity vaccine for Japanese encephalitis. CONSTITUTION:A Japanese encephalitis virus JaOArS982 strain is cultured by C6/36 cell culture originated from Aedes albopictus which is a kind of mosquito and a virus RNA is separated from supernatant liquid of the cultured product. cDNA is prepared from the virus RNA using an oligonucleotide (dGTACGGDTTCCCACATTTGGTCTCC) and cloned to a cloning vector pUC13. A clone S22 having a length of 2,500bp, containing 5'-terminal of virus RNA and containing a gene region coding V3 protein of Japanese encephalitis is separated from the cloned product. XboI linkers are linked to both terminals of the clone to obtain a clone S22X. A plasmid pJM105 for the manifestation of V3 protein gene is constructed by using a plasmid pS22X and a manifestation vector YEp133PCT.
