<p>PURPOSE:To enable the culture and maintenance of hepatic cell over a long period, by planting hepatic cell to a feeder layer of a fibroblast cell which is terminated its proliferation capability by X-ray irradiation, or by planting fibroblast cell under a porous thin film and planting hepatic cell to the opposite side of the film. CONSTITUTION:3T3 terminated its proliferation capability by the X-ray irradiation with conventional X-ray radiation apparatus at a dose of 3,000-10,000 rad, fibroblast cell originated from liver, fibroblast cell originated from skin, etc., is used as a feeder layer. Hepatic cell originated from mammal is subjected, immediately after separation, to stacking culture on the feeder layer to enable the long-term maintenance of hepatic cell. As an alternative method, a collagen solution is formed in the form of a sheet, fibrillated and crosslinked by UV irradiation to obtain active collagen thin film. A fibroblast cell is planted under the collagen film and a hepatic cell is planted to the reverse side of the film. The cell can be cultured over a long period in a culture liquid such as Green medium. It is also possible to exclusively separate and recover the hepatic cell.</p> |
