PURPOSE:To prevent the lowering of plasminogen activity in the heat-treatment for the inactivation of contaminative viruses, by heating a plasminogen- containing aqueous solution in the presence of saccharose and aprotinin. CONSTITUTION:A plasminogen-containing aqueous solution originated from human plasma and human placenta is heated in the presence of saccharose and aprotinin to inactivate contaminative viruses (e.g. hepatitis virus). Similar to various other blood preparations, it is necessary to carry out the heat- treatment of a plaminogen preparation e.g. at 60 deg.C for 10hr to inactivate contam inative viruses, however, major part of plasminogen is also inactivated by con ventional heat-treatment. The inactivation of plasminogen in heat-treatment can be prevented by adding sucrose to the plasminogen-containing aqueous solution at a final concentration of 0.5g/ml - saturated concentration and aprotinin at a final concentration of >=1.5 KIU/ml. |
