PURPOSE:To purify histidine, by subjecting an aqueous solution containing impurities consisting essentially of acidic amino acids, sulfate radicals, chlorine ions, coloring matters, etc., to ion exclusion chromatography using a strong acidic cation exchange resin. CONSTITUTION:An aqueous solution containing impurities consisting essentially of acidic amino acids, sulfate radicals, chlorine ions, coloring matters, etc., is adjusted to the isoelectric point (pH=7.47) of the histidine and then injected into the upper part of a column filled with a strong acidic cation exchange resin. Water is then passed through the column to initially elute the contaminating impurities and then the histidine. Examples of the histidine solution include histidine fermentation solution, histidine germ-free fermentation solution, dissolved solution of crude histidine crystals, histidine crystallization mother liquor, etc. The strong acidic cation exchange resin must be a type corresponding to the cation contained in the solution to be treated. In the case of the histidine fermentation solution, the cation exchange resin is used in the form a of an ammonium salt. |
